Description

From QIAGEN, this kit is designed for the simultaneous removal of human/mouse/rat and bacterial ribosomal RNA during NGS library preparation, supporting epidemiological research and dual RNA-seq workflows. The technology works by inhibiting reverse transcription of unwanted rRNA, even from challenging inputs such as formalin-fixed paraffin-embedded (FFPE) samples. The inline protocol takes approximately one hour and involves combining the FastSelect reagent with the RNA sample, optional heat fragmentation, a controlled cool-down from 75°C to 25°C over 14 min, a bead-based cleanup, and continuation into first-strand synthesis.

The kit has been tested for compatibility with stranded RNA library prep kits from QIAGEN, Illumina, New England Biolabs, and Roche, and has been verified on total RNA isolated from single-species bacterial clones, complex meta-bacterial samples, cells, fresh/frozen tissue, FFPE tissue, whole blood, and serum/plasma (including exosomes). For the QIAseq Stranded Total RNA Lib Kit, the total RNA input range is 100 ng – 1 µg. Fragmentation conditions are provided for input RNA across a range of RIN values, including ≥8 (e.g., 89°C for 8 min for ~175–225 bp mean inserts) down to ≤2 (no fragmentation).

Upon receipt, the FastSelect -rRNA HMR and FastSelect 5S/16S/23S components are stored at −30 to −15°C, while the remaining components (Nuclease-free Water, FastSelect FH Buffer, and QIAseq Beads) are stored at 2–8°C; QIAseq Beads should not be frozen. The kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.
