QIAGEN delivers sensitive targeted sequencing for somatic variant detection through its QIAseq portfolio, which uses Unique Molecular Index (UMI) tagging to correct PCR and sequencing errors before bioinformatics analysis. The 12-sample kit format processes 12 indexed libraries in parallel starting from 10 ng of fragmented genomic DNA, including material extracted from formalin-fixed paraffin-embedded (FFPE) tissue sections or low-input plasma samples for liquid biopsy research.

Library construction follows a streamlined single-day workflow: enzymatic fragmentation and end repair, adapter ligation with molecular barcodes, hybrid-capture enrichment using biotinylated RNA baits at 65°C for 2 hours, streptavidin bead pull-down, and PCR amplification with sample-indexing primers compatible with Illumina sequencers. Final libraries are quantified via qPCR or QIAseq Library Quantifier assays before pooling for paired-end 150 bp sequencing on MiSeq, NextSeq, or NovaSeq instruments.

Variant calling sensitivity reaches 1% allele frequency with the UMI-aware pipeline, supporting tumor-normal comparison, minimal residual disease monitoring, and cancer hotspot profiling studies. Capture probes tile across hotspot exons and full coding regions of clinically actionable oncology genes. All reaction components ship in single-use tubes and should be stored at -20°C, with a 12-month shelf life from manufacture. The kit is for research use only in translational genomics laboratories.
