Targeted enrichment of selected genomic regions for next-generation sequencing is enabled through QIAGEN's QIAseq DNA Custom panel (96), built around Single Primer Extension (SPE) chemistry and Unique Molecular Index (UMI) molecular barcoding. The panel supports combinatorial sample indexing for up to 96 samples per run, dramatically increasing throughput while preserving low allele-frequency detection sensitivity. The wet-lab workflow accommodates small amounts of fragmented input DNA, including material recovered from formalin-fixed paraffin-embedded (FFPE) tissue, and produces libraries compatible with Illumina sequencing platforms.

Custom design projects are configured through QIAGEN's GeneGlobe portal, where researchers submit target regions, define coverage requirements, and receive a panel optimized for their specific research application. Common use cases include somatic variant detection in tumor-normal pairs, identification of low-frequency variants below 1% allele frequency, SNP genotyping, characterization of gene panels for hereditary disease research, and pathogen surveillance studies. Integrated UMI-based error correction enables reliable discrimination of true low-frequency variants from sequencing artifacts, which is particularly valuable when working with degraded or low-input clinical samples.

Each kit includes all reagents required for the two-tube library preparation protocol, including adapters, amplification primers, and cleanup beads. Store all components at -20 °C to preserve enzymatic activity, and avoid repeated freeze-thaw cycles of master mix components prior to PCR amplification and indexing. The product is intended for research use only and supports in vitro analytical workflows within genomics and translational research laboratories.
