QIAseq DIRECT SARS-CoV-2 Kit H

A targeted viral RNA library preparation kit from QIAGEN, designed for SARS-CoV-2 and compatible with Illumina sequencers. The workflow provides an approximately five-hour turnaround time from viral RNA sample to sequencing, combining cDNA synthesis, SARS-CoV-2 enrichment, and library amplification and indexing reagents. The Enhanced protocol removes the need for normalization and quantification prior to amplification and indexing, with an RNA input of 5 μl used regardless of viral load in the sample extract.

Amplification uses two primer pools that generate overlapping amplicons of approximately 250 bp, removing the need for fragmentation prior to sequencing on Illumina platforms. The kit provides 10 bp unique dual indexes (UDIs), and with a paired-end cycle read length of 149, the sequencing time is just over 28 hours.

Use of the Enhanced protocol requires the QIAseq DIRECT SARS-CoV-2 Enhancer, purchased separately, and an optional QIAseq DIRECT Booster Panel A is available to increase coverage across multiple regions of the SARS-CoV-2 genome and help overcome S Gene Target Failure (SGTF).
