Utilizing a novel combined end-repair and ligation reaction, the kit streamlines the entire NGS library preparation process to 30 minutes through a one-tube reaction. The full procedure can be performed at room temperature, enabling automation on instruments lacking a thermo-block. The one-tube protocol removes the need to transfer reagents, increasing efficiency and more effectively capturing insert amplicons, while also reducing the risk of contamination or sample mix-up that can occur with manual processing.

The procedure is optionally PCR-free to avoid introducing sequence duplicates or PCR bias, generating high-quality libraries optimized for sequencing on any Illumina sequencing instrument from a range of input materials. It supports the construction of Illumina-compatible libraries from multiplexed PCR amplicons.
