Description

QIAGEN's targeted sequencing panel for metagenomic analysis enables high-throughput next-generation sequencing of microbial communities. Using a two-stage PCR workflow, the QIAseq 16S/ITS Region Panel enriches conserved regions of the ribosomal 16S gene (for archaea and bacteria) and the ITS gene (for fungi), enabling taxonomic classification of bacterial and fungal populations in a sample. To address the common problem of reagent contamination that can distort low-abundance community profiles, the kit includes an ultra-clean production (UCP) Master Mix and UCP PCR Water that are manufactured and quality controlled in a clean environment.

To counter the low base-diversity challenge inherent to single-amplicon 16S sequencing, the panel incorporates phased primers that add additional bases before the 16S primer, increasing base diversity during the early sequencing cycles and supporting higher Q30 quality scores. The workflow includes a bead-based cleanup after the first PCR, followed by library amplification that introduces sample indices using QIAseq Index Kits (sold separately). Final libraries are quality controlled using Bioanalyzer or TapeStation instruments and quantified with the QIAseq Library Quant system before sequencing on Illumina MiSeq platforms. The kit is compatible with the QIAseq 16S/ITS Smart Control, a process control that monitors library construction and tracks environmental contamination introduced during processing.

The kit contains primer pools, UCP Master Mix, UCP PCR Water, QIAseq Beads, and a Quick-Start Protocol. Each lot is tested against predetermined specifications under QIAGEN's ISO-certified Quality Management System, and the products are intended for molecular biology applications. They are not intended for the diagnosis, prevention, or treatment of disease.
