QIAquick PCR Purification Kit (250)

This silica-membrane spin-column kit from QIAGEN provides buffers and collection tubes for purifying PCR products greater than 100 bp, with a binding capacity of 10 µg. Using a simple bind-wash-elute procedure with an elution volume of 30–50 μl, the system recovers DNA fragments from 100 bp to 10 kb. An optional pH indicator helps determine the optimal binding pH for each sample.

The procedure removes primers, nucleotides, enzymes, mineral oil, salts, agarose, and ethidium bromide from DNA samples. Spin columns fit a conventional table-top microcentrifuge or any vacuum manifold with luer connectors such as QIAvac 24 Plus with QIAvac Luer Adapters, and the workflow can be fully automated on the QIAcube Connect. Standard protocols can also be run on the TRACKMAN Connected system paired with PIPETMAN M Connected pipettes from Gilson. GelPilot Loading Dye is supplied, containing xylene cyanol, bromophenol blue, and orange G tracking dyes.

According to the QIAGEN quick-start protocol, the kit can be stored at room temperature (15–25°C) for up to 12 months. Buffer PB is added at 5 volumes to 1 volume of the PCR reaction, and DNA is eluted in Buffer EB (10 mM Tris·Cl, pH 8.5) or water (pH 7.0–8.5), with centrifugation at 17,900 x g (13,000 rpm). Recovered DNA is ready for sequencing, microarray analysis, ligation and transformation, restriction digestion, labeling, microinjection, PCR, and in vitro transcription.
