QIAquick PCR & Gel Cleanup Kit (100)

A silica-membrane-based purification system from QIAGEN, the QIAquick PCR & Gel Cleanup Kit provides spin columns, buffers, and collection tubes for purification of DNA fragments from enzymatic reactions and agarose gels. The system uses a simple bind-wash-elute procedure that removes primers, nucleotides, enzymes, mineral oil, salts, agarose, ethidium bromide, and other impurities from DNA samples.

Reported specifications include a binding capacity of 10 µg, an elution volume of 30–50 µl, and up to 80% recovery of DNA. Sources cite different size ranges, with one indicating purification of PCR products >100 bp and DNA of up to 10 kb, and another reporting a fragment size range of 70 bp – 10 kb. Gel slices of up to 400 mg can be processed using a microcentrifuge or vacuum manifold, with throughput of 1–24 samples. The kit is suitable for use with standard or low-melt agarose gels in TAE or TBE buffer.

An optional pH indicator helps determine the optimal pH for DNA binding, and GelPilot Loading Dye containing xylene cyanol, bromophenol blue, and orange G is provided for sample processing. For optimal results, the kit is recommended for use with the QIAvac 24 Plus, and the procedure can be fully automated on the QIAcube Connect. Standard protocols can also be executed using the TRACKMAN Connected system with Bluetooth-enabled PIPETMAN M Connected pipettes. Purified DNA is ready for direct use in sequencing, ligation and transformation, restriction digestion, labeling, microinjection, PCR, and in vitro transcription.
