QIAquick Gel Extraction Kit (250)

Designed for laboratory DNA purification workflows, this QIAGEN kit enables extraction of DNA fragments ranging from 70 bp to 10 kb from standard or low-melt agarose gels in TAE (Tris·acetate/EDTA) or TBE (Tris·borate/EDTA) buffer, and also supports DNA cleanup from enzymatic reactions. The QIAquick Gel Extraction Kit is suitable for purification of up to 10 μg of DNA and falls within QIAGEN's Secondary Sample Technologies consumables range.

Processing is carried out by centrifugation at 17,900 x g (13,000 rpm) at room temperature (15–25°C). The maximum amount of gel per column is 400 mg (about 0.4 cm³). For dissolution, 3 volumes of Buffer QG are added per 1 volume of gel (100 mg gel ~ 100 μL), or 6 volumes of Buffer QG for agarose gels above 2%. DNA is bound to a QIAquick spin column with a maximum loading volume of 800 μL per spin. For purifying fragments ≥500 bp or ≤4 kb, 1 volume of isopropanol is added to increase yield.

The wash step uses 750 μL of Buffer PE, and elution is performed with 50 μL of Buffer EB (10 mM Tris·Cl, pH 8.5) or water (pH 7.0–8.5), yielding an average eluate volume of 48 μL. For higher DNA concentration, 30 μL of elution buffer can be used, producing an average eluate volume of 28 μL. Maximum elution efficiency is achieved between pH 7.0 and 8.5.
