QIAprep Spin Miniprep Kit (50)

A spin column-based plasmid purification system from QIAGEN designed for isolating high-purity plasmid or cosmid DNA. The kit uses silica membrane technology, where the QIAprep 2.0 Spin Columns bind up to 20 µg DNA in the presence of a high concentration of chaotropic salt, allowing elution in a small volume of low-salt buffer. Yields of up to 30 µg can be obtained using the High-Yield Supplementary Protocol.

The kit follows a simple bind-wash-elute workflow. Bacterial cultures are lysed and cleared by centrifugation, then applied to the QIAprep 2.0 module where plasmid DNA adsorbs to the silica membrane. Impurities are washed away and pure DNA is eluted. The minimum elution volume is 50 µl and the column reservoir capacity is 800 µl. Recommended culture volumes are 1–5 ml for high-copy plasmids and 1–10 ml for low-copy plasmids or cosmids. Twenty-four minipreps can be completed in under 30 minutes, with throughput of 1–24 samples per run.

Processing is manual by vacuum or microcentrifugation, or fully automated on the QIAcube Connect. QIAprep 2.0 Spin Columns can be vacuum processed using the QIAvac 24 Plus or any commercial manifold with luer connectors. The kit includes GelPilot Loading Dye, which contains three tracking dyes (xylene cyanol, bromophenol blue, and orange G) for agarose gel analysis. Eluted DNA is immediately ready to use for fluorescent and radioactive sequencing (including capillary sequencing), ligation, cloning, transformation, PCR, and restriction digestion. In addition to Escherichia coli, plasmid DNA can be purified from Saccharomyces cerevisiae, Bacillus subtilis, and Agrobacterium tumefaciens.
