QIAPrep&amp CRISPR (250x)

From QIAGEN, this kit is designed for characterization of CRISPR editing events, enabling rapid processing of cultured cells and fast amplification of a genomic region of interest. The lysate can be used directly for amplification of the target region without the need for additional purification.

Intended for research use, it is optimized for adherent and suspension-cultured cells derived from human, mouse, and rat. The kit should be stored at −30 to −15°C in a constant-temperature freezer; after first thawing, the Cell Lysis Buffer is stored at 2–8°C. Key components include AllTaq PCR Master Mix (4x), CRISPR-Q Custom PCR Assay (20x), an optional Master Mix Tracer supplied as a 125x concentrate, and Q-Solution to facilitate amplification of templates that have a high degree of secondary structure or that are GC-rich.

The quick-start protocol describes a PCR cycling program with an initial activation step at 95°C, followed by 40 cycles of 3-step cycling — denaturation at 95°C, annealing at 60°C, and extension at 72°C — and a final extension at 72°C, in a total reaction volume of 20 µl. Reactions containing the Master Mix Tracer can be loaded directly onto agarose gels after cycling.
