QIAGEN Plasmid Plus Maxi Kit (100)

Designed for ultrafast, large-scale plasmid DNA isolation, this QIAGEN kit enables purification of up to 10 mg of highly pure plasmid DNA from bacterial cultures. A vacuum-based workflow allows up to 24 samples to be processed in parallel, reducing hands-on time. Low elution volumes deliver highly concentrated plasmid DNA for direct use without ethanol precipitation, and a dedicated wash buffer reduces endotoxin levels. The purified DNA is suitable for PCR, cloning, sequencing, and transfection into sensitive cell lines.

Based on anion-exchange silica membrane technology, the kit is stored at room temperature (15–25°C) for up to 24 months. According to the manufacturer's protocol, recommended LB culture volumes are 80–100 ml for high-copy plasmids or up to 200 ml for low-copy plasmids using the standard protocol, and 100–130 ml for high-copy plasmids using the high-yield protocol.

Key protocol steps include lysing bacterial pellets with Buffer P1 and Buffer P2, clearing the lysate with Buffer S3 through a QIAfilter cartridge, binding DNA on a QIAGEN Plasmid Plus spin column under approximately −300 mbar vacuum, washing with 0.7 ml Buffer ETR and 0.7 ml Buffer PE, and eluting with 400 μl Buffer EB or water.
