HiSpeed Plasmid Midi Kit (25)

A QIAGEN anion-exchange-based kit designed for fast, large-scale purification of transfection-grade plasmid or cosmid DNA without centrifugation. The kit contains QIAfilter Cartridges, HiSpeed Tips, and QIAprecipitator modules that replace centrifugation steps in the classic anion-exchange procedure, enabling syringe-format processing without a vacuum manifold.

Suitable for high-copy plasmids and cosmid DNA from 50 ml – 150 ml culture volumes, with a single sample processed per run in approximately 45 minutes. Up to 200 µg of high-copy plasmid DNA can be obtained, with yields depending on plasmid copy number, insert size, host strain, and culture medium. Intended applications include transfection, cloning, sequencing, and gene silencing.

The QIAfilter Cartridge clears bacterial lysates and removes SDS precipitates in seconds, while HiSpeed Tips operate by gravity flow without running dry. The proprietary anion-exchange resin delivers DNA purity equivalent or superior to two successive rounds of CsCl gradient centrifugation. Eluted DNA is precipitated with isopropanol and concentrated using the QIAprecipitator module, which traps the precipitated DNA and eliminates the risk of pellet loss associated with decanting; the final DNA is eluted directly into a microcentrifuge tube with TE buffer or water.
