Reverse transcription and PCR amplification are consolidated into a single closed tube by this QIAGEN 25-reaction kit, eliminating the need to open vessels between cDNA synthesis and thermal cycling. The kit ships with a proprietary enzyme blend that pairs HotStarTaq DNA Polymerase with Sensiscript Reverse Transcriptase, along with a specially formulated OneStep buffer, a dNTP mix, and RNase-free water. Hot-start activation keeps the polymerase inactive until the initial incubation step, suppressing non-specific amplification and improving yield from low-copy transcripts.

Each 50 µL reaction accommodates up to 1 µg of total RNA, making the system well suited to gene expression analysis, validation of microarray data, detection of viral RNA, and characterization of rare splice variants. Built-in RNase inhibitor activity within the enzyme mix protects template integrity during setup, while the integrated workflow shortens hands-on time and reduces contamination risk compared with two-step protocols.

Store the kit at -20°C and protect all components from repeated freeze-thaw cycles; aliquot the enzyme mix upon first use if frequent small-volume setup is anticipated. The kit is intended for research use only and is supplied in a single box containing all reagents required for 25 reactions.
