QIAGEN Multiplex PCR Kit (1000)

Designed by QIAGEN for fast and efficient multiplex PCR without lengthy optimization, this system enables amplification of two or more DNA products in parallel within a single reaction tube. It is widely used for genotyping applications and DNA testing in research, forensic, and diagnostic laboratories. The kit provides high specificity through a built-in hot start: HotStarTaq DNA Polymerase is supplied in an inactive state at ambient temperatures and is activated by a 15-minute, 95°C incubation step, preventing the formation of misprimed products and primer-dimers during reaction setup and the first denaturation step.

At the core is a 2x Multiplex PCR Master Mix containing HotStarTaq DNA Polymerase—a modified form of a recombinant 94 kDa DNA polymerase originally isolated from Thermus aquaticus and cloned in E. coli (EC 2.7.7.7)—together with the QIAGEN Multiplex PCR Buffer containing 6 mM MgCl2 at pH 8.7 (20°C), an ultrapure dNTP mix, and the synthetic factor MP that supports balanced primer annealing and extension irrespective of primer sequence. Q-Solution is supplied as a 5x concentrate to facilitate amplification of GC-rich templates or those with a high degree of secondary structure, and RNase-free water is included for reaction setup.

QIAGEN recommends primer pairs with a Tm greater than 68°C used at equal concentrations of 0.2 µM each. The kit supports protocols for standard multiplex PCR with amplicons up to 1.5 kb and for microsatellite loci or small amplicons up to 0.5 kb. Quality control testing covers PCR performance, reproducibility, and sensitivity in parallel multiplex reactions. The product is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of disease.
