Description

QIAGEN's silica-based gel extraction system purifies DNA fragments of 40 bp to 50 kb from TAE or TBE agarose gels, polyacrylamide gels, and solutions without phenol extraction or ethanol precipitation. The QIAEX II Gel Extraction Kit uses silica-gel particles in a manual tube format with a binding capacity of 5 µg per 10 µl of suspension and a minimum elution volume of 20 µl of Tris buffer or water.

Recovery varies with fragment size, reaching 75% at 44 bp, 75% at 75 bp, 95% at 500 bp, 85% at 7.5 kb, 75% at 23.5 kb, and 60% at 48.5 kb, with an overall recovery range of 60–95%. The system removes <10mers, 17–40mers dye terminators, and proteins. The solubilization and binding buffer contains a pH indicator that signals whether the mixture is at the optimal pH ≤7.5 for DNA adsorption; if the pH is too high, it can be adjusted by adding 10 µl of 3 M sodium acetate (pH 5.0). The batch procedure can be scaled up to bind 15 µg of DNA using 30 µl of suspension.

DNA purified with this QIAGEN kit is suitable for downstream molecular biology applications, including restriction digestion, labeling, ligation, PCR, sequencing, and microinjection. The kit is intended for molecular biology use and should be stored dry at room temperature (15–25°C), where it remains stable for at least 12 months without reduction in performance, capacity, or quality of separation.
