QIAGEN provides a spin-column kit designed for the simultaneous purification of viral RNA and DNA from plasma, serum, and other cell-free body fluids. The procedure combines the selective binding properties of a silica-based membrane with flexible elution volumes, and processed viral nucleic acids are eluted free of proteins, nucleases, and other impurities.

The kit operates through a simple four-step process — lyse, bind, wash, and elute — carried out using MinElute columns in a standard microcentrifuge or fully automated on the QIAcube Connect system. The spin procedure is optimized for a starting sample volume of 200 µl, with elution volumes adjustable between 20 and 150 µl. Yields of viral nucleic acid isolated from biological samples are normally below 1 µg, and quantitative amplification methods are recommended for determination of yields.

Lysis is performed under highly denaturing conditions at elevated temperatures in the presence of QIAGEN Protease and Buffer AL, ensuring inactivation of RNases. Binding conditions are adjusted by adding ethanol to allow optimal binding of viral RNA and DNA to the silica-gel membrane, while protein and other contaminants that can inhibit downstream enzymatic reactions are removed during three wash steps. The procedure is designed to prevent sample-to-sample cross-contamination, allows safe handling of potentially infectious samples, and delivers pure nucleic acid in less than one hour.
