Description

A vacuum-based purification system from QIAGEN enables high-yield DNA purification from transport media using MinElute spin columns and low-volume elution. The QIAamp MinElute Media Kit provides a convenient procedure for purification of nucleic acids from liquid media, such as cervical swab transport media, including PreservCyt or SurePath solution. Samples of 250 µl can be processed in less than 90 minutes, with DNA eluted in 20–150 µl of Buffer AVE, ready for use in amplification reactions or storage.

The kit combines the selective binding properties of a silica-based membrane with flexible elution volumes and supports purification of cellular, bacterial, and viral nucleic acids for applications such as PCR and real-time PCR. The procedure comprises four steps (lyse, bind, wash, elute) and is carried out on QIAvac 24 or QIAvac 24 Plus vacuum manifolds in batches of 24 samples, with no detectable sample-to-sample cross-contamination and safe handling of potentially infectious samples. Purification can also be automated on the QIAcube Connect.

MinElute spin columns should be stored at 2–8°C upon arrival; short-term storage (up to 4 weeks) at 15–25°C does not affect performance, though storage temperature should not exceed 25°C for optimal quality. Purified nucleic acids are free of proteins, nucleases, and other impurities. Fluorometric measurements or quantitative RT-PCR and PCR are recommended over spectrophotometric methods for quantifying low amounts of RNA or DNA.
