Description

A tri-substituted purine derivative supplied by STEMCELL Technologies as a Hedgehog pathway activator. The compound directly binds to and activates the Hedgehog receptor Smoothened (SMO), with an EC50 of 1 µM (Sinha and Chen). It is provided as a crystalline solid with a chemical purity of ≥ 98%.

The chemical is 9-cyclohexyl-N-[4-(morpholinyl)phenyl]-2-(1-naphthalenyloxy)-9H-purin-6-amine (CAS 483367-10-8), with molecular formula C31H32N6O2 and molecular weight 520.6 g/mol. It is soluble in DMSO at ≤ 20 mM; for example, a 10 mM stock in DMSO can be prepared by resuspending 1 mg in 192 µL of fresh DMSO. Stock solutions should be prepared fresh, stored at -20°C, and aliquoted to avoid repeated freeze-thaw cycles. The product is stable at -20°C as supplied for 12 months from date of receipt and should be protected from prolonged exposure to light. The compound has low solubility in aqueous media; for cell culture use, stock solution should be diluted into culture medium immediately before use, and the final DMSO concentration should be kept below 0.1% to avoid potential cell toxicity.

Published studies report that the compound promotes differentiation of ventral spinal progenitor cells and motor neurons from human pluripotent stem cells, promotes osteoblast differentiation from human and mouse mesenchymal cells, and inhibits differentiation and maturation of adipocytes from human mesenchymal cells. It is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS) and is intended for research use only.
