Description

A small molecule from STEMCELL Technologies, this tri-substituted purine derivative activates the Hedgehog signaling pathway by directly binding to and activating the Hedgehog receptor Smoothened (SMO), with an EC₅₀ of 1 µM (Sinha and Chen).

The chemical name is 9-cyclohexyl-N-[4-(morpholinyl)phenyl]-2-(1-naphthalenyloxy)-9H-purin-6-amine. Purmorphamine is supplied as a crystalline solid with a purity of ≥98%, a molecular weight of 520.6 g/mol, and the chemical formula C₃₁H₃₂N₆O₂ (CAS 483367-10-8). The compound has low solubility in aqueous media. Reported solubilities include ~10 mg/mL in DMSO, ~20 mg/mL in DMF, and ~0.25 mg/mL in a 1:2 solution of DMF:PBS (pH 7.2). It is stable for 12 months from date of receipt when stored at -20°C and protected from prolonged exposure to light. For cell culture applications, stock solutions should be diluted into culture medium immediately before use, and final DMSO concentration should be kept at or below 0.1% to avoid potential cell toxicity.

Published research applications include promoting differentiation of ventral spinal progenitor cells and motor neurons from human pluripotent stem cells, promoting differentiation of osteoblasts from human and mouse mesenchymal cells, and inhibiting differentiation and maturation of adipocytes from human mesenchymal cells.
