Puregene Tissue Kit (4 g)

This manual genomic DNA purification kit from QIAGEN uses a modified salting-out precipitation method to recover high-molecular-weight DNA from tissue samples, including fixed and paraffin-embedded tissue. The scalable procedure is suited to biobanking, genotyping, and workflows that demand high DNA integrity.

Each preparation processes 5–100 mg of tissue and yields genomic DNA in an elution volume of 50–250 µl. Cells are lysed with an anionic detergent in the presence of a DNA stabilizer; RNA is then removed by treatment with an RNA-digesting enzyme, and other contaminants such as proteins are removed by salt precipitation. Genomic DNA is recovered by alcohol precipitation and dissolved in hydration solution (1 mM EDTA, 10 mM Tris·Cl pH 7.5).

Typical run time is 25–60 minutes per prep, including cell lysis and DNA rehydration, with convenient stopping points for safe temporary storage of partially purified samples. Purified DNA typically shows an A260/A280 ratio between 1.7 and 1.9 and reaches sizes of up to 200 kb, supporting downstream applications such as PCR, restriction digestion, Southern analysis, and SNP analysis, as well as long-term archiving. The DNA can be stored at 2–8°C, –20°C, or –80°C. The kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.
