Puregene Tissue Kit (33 g)

QIAGEN's Puregene Tissue Kit enables manual purification of high-molecular-weight genomic, mitochondrial, or viral DNA from tissue samples, including fixed and paraffin-embedded tissue. The kit uses a modified salting-out precipitation method in a scalable format, with processing completed in 25–60 minutes including cell lysis and DNA rehydration.

The procedure lyses cells with an anionic detergent in the presence of a DNA stabilizer that limits DNase activity, then removes RNA using an RNA digesting enzyme. Other contaminants such as proteins are removed by salt precipitation, and genomic DNA is recovered by alcohol precipitation and dissolved in hydration solution (1 mM EDTA, 10 mM Tris·Cl pH 7.5). Purified DNA typically exhibits an A260/A280 ratio between 1.7 and 1.9 and reaches sizes of 100–200 kb, making it well suited for biobanking, genotyping, and other archiving applications. DNA can be safely stored at 2–8°C, –20°C, or –80°C.

The kit supports sample amounts of 5–100 mg with an elution volume of 50–250 µl. Downstream applications include PCR, restriction digest, Southern analysis, and SNP analysis. Each kit is produced under QIAGEN's ISO-certified Quality Management System, with every lot tested against predetermined specifications. Reagents provided include Cell Lysis Solution, Protein Precipitation Solution, DNA Hydration Solution, RNase A Solution, and Proteinase K. The product is intended for molecular biology applications and is not for the diagnosis, prevention, or treatment of a disease.
