Description

QIAGEN's Puregene Kits provide flexible, manual genomic DNA purification, ideal for biobanking, genotyping, and workflows needing high DNA integrity. The Puregene Cell Kit is intended for cultured cell preparations and uses a modified salting-out precipitation method that removes contaminants and enzyme inhibitors, enabling purification of highly stable DNA well-suited for archiving.

Cells are lysed with an anionic detergent in the presence of a DNA stabilizer that limits DNase activity, and RNA is removed by treatment with an RNA-digesting enzyme. Other contaminants such as proteins are removed by salt precipitation, and the genomic DNA is recovered by alcohol precipitation and dissolved in hydration solution (1 mM EDTA, 10 mM Tris·Cl pH 7.5). The scalable workflow is fully manual, requires no mixing or dilution of solutions, and provides convenient stopping points for safe temporary storage of partially purified samples.

For cultured cell preparations, the kit accepts 1 x 10^6 – 2.2 x 10^7 cells per prep, with an elution volume of 50–250 µl (varies) and an expected yield of 7 µg per 1 million cells. Per-prep time is 25–60 minutes including DNA rehydration. Purified DNA typically exhibits an A260/A280 ratio between 1.7 and 1.9, is up to 200 kb in size, and performs well in sensitive downstream applications including PCR and restriction digestion. The DNA can be safely stored at 2–8°C, –20°C, or –80°C, and is suited for PCR, restriction digest, Southern analysis, and SNP analysis.
