Puregene Cell Kit (6.7 x 109)

A scalable manual genomic DNA purification kit from QIAGEN, intended for molecular biology applications and not for the diagnosis, prevention, or treatment of a disease. Designed for cultured cells and based on a modified salting-out precipitation method, the kit supports downstream applications including PCR, restriction digest, Southern analysis, and SNP analysis, and is well-suited for biobanking, genotyping, and workflows needing high DNA integrity.

Cells are lysed with an anionic detergent in the presence of a DNA stabilizer, RNA is removed by treatment with an RNA-digesting enzyme, and other contaminants such as proteins are removed by salt precipitation. Genomic DNA is then recovered by alcohol precipitation and dissolved in hydration solution (1 mM EDTA, 10 mM Tris·Cl pH 7.5). Purified DNA typically has an A260/A280 ratio between 1.7 and 1.9 and can be up to 200 kb in size, with a reported yield of 7 µg per 1 million cells. The procedure takes 25–60 minutes plus DNA rehydration, with an elution volume of 50–250 µl (varies). Recommended sample amounts range from 1 x 10^6 to 2.2 x 10^7 cells, and purified DNA can be stored at 2–8°C, –20°C, or –80°C.

The procedure enables purification of high-molecular-weight (100–200 kb) DNA suitable for archiving, removing contaminants and enzyme inhibitors while providing convenient stopping points for safe temporary storage of partially purified samples.
