Puregene Blood Kit (120 ml)

QIAGEN's Puregene Blood Kit supports flexible, manual purification of genomic DNA from whole blood, buffy coat and body fluid, and is intended for workflows such as biobanking and genotyping that demand high DNA integrity. The scalable procedure is based on a modified salting-out precipitation method, in which cells are lysed with an anionic detergent in the presence of a DNA stabilizer that limits DNase activity, RNA is removed with an RNA-digesting enzyme, and proteins are eliminated by salt precipitation. Genomic DNA is then recovered by alcohol precipitation and dissolved in hydration solution (1 mM EDTA, 10 mM Tris·Cl pH 7.5).

Purified DNA is high-molecular-weight (100–200 kb), with an A260/A280 ratio between 1.7 and 1.9, and is suitable for archiving as well as for sensitive downstream applications including PCR, restriction digest, Southern analysis and SNP analysis. The resulting DNA can be safely stored at 2–8°C, –20°C or –80°C.

Acceptable sample inputs are 300 µl – 10 ml of whole blood, 1 x 10^7 cells, or 50 µl – 1000 µl of body fluid, with an elution volume of 250 µl – 1000 µl, a yield of 35 µg per ml of whole blood, and a preparation time of 25–60 minutes including DNA rehydration.
