Puregene Blood Kit (1000 ml)

For flexible, manual purification of high-molecular-weight genomic DNA from whole blood, buffy coat, and body fluid, the QIAGEN Puregene Blood Kit supports workflows in biobanking, genotyping, and other applications needing high DNA integrity. Using a modified salting-out precipitation method in a scalable format, the kit yields 35 µg of DNA per ml of whole blood. Sample volumes range from 300 µl to 10 ml, elution volumes from 250 µl to 1000 µl, and processing time is 25–60 minutes per preparation (plus DNA rehydration).

The procedure includes RBC Lysis Solution along with reagents to lyse cells with an anionic detergent in the presence of a DNA stabilizer that limits DNase activity. RNA is removed by an RNA-digesting enzyme, proteins and other contaminants are removed by salt precipitation, and the genomic DNA is recovered by alcohol precipitation before dissolution in hydration solution (1 mM EDTA, 10 mM Tris·Cl pH 7.5). No mixing or dilution of solutions is required, and the procedure provides convenient stopping points for safe, temporary storage of partially purified samples.

Purified DNA typically displays an A260/A280 ratio between 1.7 and 1.9 and reaches sizes up to 200 kb, making it well-suited for archiving as a long-term reference. It performs reliably in sensitive downstream applications, including PCR, restriction digestion, Southern analysis, and SNP analysis, and can be safely stored at 2–8°C, –20°C, or –80°C.
