Puregene Blood Kit (1000 ml)

QIAGEN offers the Puregene Blood Kit for flexible, manual purification of high-molecular-weight genomic DNA from whole blood, buffy coat, and body fluid samples, suited to biobanking, genotyping, and workflows that demand high DNA integrity. The scalable procedure relies on a modified salting-out precipitation method: cells are lysed with an anionic detergent in the presence of a DNA stabilizer that limits DNase activity, RNA is removed by an RNA-digesting enzyme, proteins and other contaminants are eliminated by salt precipitation, and the genomic DNA is recovered by alcohol precipitation and dissolved in hydration solution (1 mM EDTA, 10 mM Tris·Cl pH 7.5).

Purified DNA typically shows an A260/A280 ratio between 1.7 and 1.9, reaches a molecular weight of 100–200 kb (up to 200 kb in size), and can be stored at 2–8°C, –20°C, or –80°C. The kit supports an elution volume of 250 µl–1000 µl and a yield of 35 µg per ml of whole blood, accommodating sample amounts of 300 µl–10 ml of whole blood, 50 µl–1000 µl of body fluid, or up to 1 x 10⁷ cells, with each prep taking 25–60 minutes plus DNA rehydration. The resulting DNA is suitable for sensitive downstream applications such as PCR, restriction digestion, Southern analysis, and SNP analysis.

Kit components include RBC Lysis Solution, Cell Lysis Solution, Protein Precipitation Solution, DNA Hydration Solution, and RNase A Solution. Buffers and reagents are stored dry at the temperature indicated on the kit label; RNase A Solution must be refrigerated or frozen, and other reagents can be kept at room temperature (15–25°C). The kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of disease.
