The enzyme converts sheared or enzymatically fragmented input into blunt-ended, adenine-overhang substrates that ligate efficiently with sequencing adapters used in cluster generation on short-read sequencing platforms.

QIAGEN supplies the enzyme with a tailored reaction buffer optimized for activity across a broad DNA input range and to limit GC bias during the repair step. The reagent is provided in a single-use vial format and integrates with the fragmentation, ligation, and cleanup modules of the Universal NGS product family. Storage at -20 °C in a non-frost-free freezer is recommended; aliquoting after first thaw preserves activity through subsequent freeze-thaw cycles. The product is intended for research use only.
