Description
QIAGEN blood-collection style tubes are formulated to stabilise intracellular RNA in human bone marrow aspirates immediately at point of collection. Each tube measures 16 x 125 mm, features a paper label and a BD Hemogard red closure, and contains 11.1 ml of an RNA-stabilising additive. A 2 ml bone marrow sample, collected using standard medical practice, is transferred into the tube, where the additive lyses all cells in the sample and protects RNA from degradation by RNases and other enzymes, preserving in vivo expression profiles and halting ex vivo changes in gene expression.
Stabilization is maintained for up to 3 days at room temperature (15–25°C), up to 5 days at 2–8°C, and for long-term storage at –20°C or –70°C, with high RNA integrity numbers (RIN) measured in samples even after 5 days at 2–8°C and preservation of integrity through freeze-thaw cycles. In a study of 295 bone marrow samples, yields of RNA isolated from 2 mL human bone marrow were between 2.0 and 44.7 µg (mean 15.8 µg, median 11.0 µg).
For storage below –20°C, tubes should first be frozen for 24 hours at –20°C before transfer to –70°C, ideally in a wire rack rather than an expanded polystyrene foam tray. The tubes are intended for use with the PAXgene Bone Marrow RNA Kit for isolation and purification of intracellular RNA from human whole bone marrow samples. The product is for research use only and is not for use in diagnostic procedures or for blood collection.
