PAXgene Blood miRNA Kit (50)

QIAGEN's PAXgene Blood miRNA Kit is designed for the purification of total RNA — including miRNA, mRNA, and RNA smaller than 200 nucleotides — from whole blood. The kit uses silica-membrane technology in a spin-column format and is used in conjunction with PAXgene Blood RNA Tubes, which collect, stabilize, and transport blood and prevent changes to gene expression profiles during storage or transport at ambient temperature.

Cell lysis and RNA stabilization occur directly in the PAXgene Blood RNA Tubes upon blood collection. After removal of cellular debris, kit buffers promote RNA binding to the silica membrane, contaminants are washed away, and trace DNA is removed by treatment with DNase I. Pure RNA, including miRNA, is then eluted in a low-salt elution buffer and denatured by heating. The purified material is ready for downstream applications such as RT-PCR and real-time RT-PCR, differential display, cDNA synthesis, northern, dot, and slot blot analyses, primer extension, and microarray analysis.

Specifications include a silica-technology spin-column format, whole-blood main sample type, 2.5 ml sample amount, 80 µl elution volume, and a yield of 3–18 µg per 2.5 ml sample. Processing is manual by centrifugation or automated on QIAcube instruments; up to 12 samples can be processed in a single run, with run times of 90 min for 12 samples manually and 125 min for 12 samples automated. As part of QIAGEN's Go Greener program, the kit contains no phenol or Triton, and the thickness of the plastic blister component in its packaging has been reduced from 300 µm to 200 µm compared with previous versions of the kits.
