Description

Recombinant Sonic hedgehog (Shh) from STEMCELL Technologies is a member of the Hedgehog family of secreted signaling proteins that has a prominent role in patterning during early mammalian development. It is expressed during early embryogenesis in a variety of tissues. During patterning of the central nervous system, Shh acts directly on cells to specify neural cell fates, regulates proliferation and survival of oligodendrocyte precursors and neural crest cells, and stimulates proliferation, differentiation, and axon growth. The recombinant protein has also been reported to regulate T cell differentiation and activation.

The recombinant mouse protein is produced in E. coli and is lyophilized after dialysis against phosphate-buffered saline. It is supplied as a solid powder with a stated purity of ≥ 95% and an endotoxin level of ≤ 0.2 EU/μg protein as measured by kinetic limulus amebocyte lysate (LAL) analysis. The predicted molecular mass is 19.8 kDa (UniProt accession Q62226). Biological activity is determined by inducing alkaline phosphatase production in C3H/10T1/2 cells, with a specific activity of ≥ 1 x 10^3 units/mg (EC50 ≤ 1 μg/mL); in a representative assay using CCL-226 cells, an EC50 of 0.8 μg/mL was observed.

The product is for research use only. Vials should be centrifuged before opening and the contents resuspended in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex. Reconstituted material can be stored at 2 - 8°C for up to 2 weeks or at -20°C to -80°C for up to 3 months, and the lyophilized product is stable at -80°C for 12 months from the date of receipt. Repeated freeze-thaw cycles should be avoided.
