Description

A BMP pathway antagonist of the TGF-β family, Mouse Recombinant Noggin from STEMCELL Technologies supports maintenance of undifferentiated human embryonic stem cells cultured in mouse embryonic fibroblast (MEF)-conditioned medium and promotes dopaminergic differentiation of embryonic stem cells with subsequent survival of dopamine neurons. The protein is essential for development of ectodermal structures including neural tube, tooth, hair follicle, and eye, and influences patterning of mesodermal somites, skeletal structures, chondrogenesis, osteogenesis, and joint formation. Alternative names include NOG, SYM1, SYNS1, Symphalangism 1 (proximal), and Synostoses (multiple) syndrome. The protein is produced in CHO cells to ≥95% purity.

The lyophilized product is supplied after dialysis against phosphate-buffered saline. Reconstitute by pipetting the solution down the sides of the vial in sterile water or PBS to at least 0.1 mg/mL; do not vortex. The predicted molecular mass is 29-31 kDa and the endotoxin level is ≤0.2 EU/μg protein as measured by kinetic LAL analysis. Biological activity is determined by inhibition of BMP-4 induced alkaline phosphatase production in ATDC5 cells, with a specific activity of ≥1.67 x 10^4 units/mg (EC50 ≤60 ng/mL) in the presence of 10 ng/mL human BMP-4. Cross-reactivity is not determined.

Store the lyophilized product at -80°C; it is stable as supplied for 12 months from the date of receipt. After reconstitution, store at 2-8°C for up to 1 week or at -20°C to -80°C for up to 2 months, avoiding repeated freeze-thaw cycles. The product is intended for research use only.
