Description

From STEMCELL Technologies, this Mouse Recombinant IL-6 is a pleiotropic growth factor with biological activities spanning immune regulation, hematopoiesis, and oncogenesis. The cytokine is produced by a range of cell types, including T cells, B cells, monocytes and macrophages, fibroblasts, hepatocytes, vascular endothelial cells, and various tumor cell lines. On its own or together with factors such as IL-2 and interferon-γ, it stimulates the proliferation of B cells, T cells, and hybridoma cells. In combination with cytokines such as IL-3, GM-CSF, and SCF, it promotes hematopoietic progenitor cell proliferation and differentiation in vitro. IL-6 additionally stimulates T-cell proliferation and differentiation, osteoclast formation, and acts on hepatocytes, cardiomyocytes, and macrophages. Signaling is mediated through a cell-surface type I cytokine receptor complex comprising the ligand-binding IL-6α (CD126) chain and the signal-transducing gp130 subunit, with binding leading to activation of the JAK/STAT pathway.

Alternative names include B cell differentiation factor, BSF-2, IFN-β2, and Interleukin-6. The UniProt accession number is P08505 and the predicted molecular mass is 21.9 kDa. The protein is expressed in E. coli and supplied lyophilized from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid (TFA). Specific activity is ≥ 1 x 10^6 units/mg; the EC50 is ≤ 50 pg/mL in a B9 cell proliferation assay and ≤ 1 ng/mL in a 7TD1 cell proliferation assay. Purity is ≥ 95% and the endotoxin level is ≤ 1 EU/μg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis. Cross-reactivity is reported with rat and monkey.

Before opening, the vial should be centrifuged and the contents reconstituted in 10 mM hydrochloric acid to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex. If the reconstituted product will not be used immediately, dilution with bovine serum albumin (BSA) to a final concentration of 0.1% is recommended. Store the lyophilized product at -20 to -80°C; it is stable as supplied for 12 months from the date of receipt. After reconstitution, general guidance is to avoid storage at 2 - 8°C for more than 1 month or at -80°C for more than 3 months, and to avoid repeated freeze-thaw cycles. The product is intended for research use only and is not for human or animal diagnostic or therapeutic applications. The manufacturer's quality management system is certified to ISO 13485.
