Description

Supplied by STEMCELL Technologies as a lyophilized preparation, this recombinant mouse Interleukin 17A (IL-17A) is intended for cell culture research use. The protein is expressed in E. coli and purified to ≥ 95% purity, formulated from a sterile filtered solution containing 0.1% trifluoroacetic acid. It is a homodimer composed of two 15.1 kDa subunits, giving a total predicted molecular mass of 30.2 kDa, and is reported to show cross-reactivity with human and rat IL-17A. Alternative designations include CTLA-8, IL-17, Interleukin-17 and Interleukin-17A, with UniProt accession Q62386.

Biological activity is determined by measuring IL-6 production in NIH 3T3 fibroblasts after 48 hours of culture, with a representative EC50 of 0.3 - 1.5 ng/mL and a specific activity of ≥ 2 x 10^5 units/mg (EC50 ≤ 5 ng/mL). Endotoxin level, measured by kinetic limulus amebocyte lysate (LAL) analysis, is ≤ 1 EU/μg protein. The lyophilized product should be stored at -20°C to -80°C and is stable as supplied for 12 months from the date of receipt.

Reconstitution instructions recommend centrifuging the vial before opening and resuspending in sterile water containing 0.1% bovine serum albumin (BSA) to at least 0.1 mg/mL by pipetting the solution down the sides of the vial, without vortexing. Once reconstituted, the product can be stored at 2 - 8°C for up to 1 month or at -20°C to -80°C for up to 3 months, avoiding repeated freeze-thaw cycles; if the reconstituted product will be used immediately, BSA is not required.
