Description

STEMCELL Technologies supplies Mouse Recombinant IL-1 beta as a lyophilized powder prepared after dialysis against phosphate-buffered saline. The recombinant protein is expressed in E. coli with a purity of ≥ 95%, and the product is intended for use as a laboratory reagent for research purposes only; it is not intended for human or animal diagnostic or therapeutic uses unless otherwise stated. The supplier's quality management system is certified to ISO 13485, and the material is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS).

IL-1β is synthesized as an inactive precursor (pro-IL-1β) that is cleaved intracellularly by caspase 1 to generate the mature, secreted form. It is released by monocytes, tissue macrophages, and dendritic cells in response to infection or injury, induces expression of acute-phase proteins, promotes extravasation of inflammatory and immunocompetent cells into affected tissues, and stimulates Th17 differentiation and B cell proliferation in an IL-6-dependent manner. Alternative names listed for the protein include Catabolin, Endogenous pyrogen, EP, LAF, LEM, Leukocyte endogenous mediator, Lymphocyte-activating factor, MCF, and Mononuclear cell factor. The recombinant product shows cross-reactivity with Human, Rat, and Monkey IL-1β, carries the UniProt accession number P10749, and has a predicted molecular mass of 17.5 kDa.

Biological activity is measured by a cell proliferation assay using D10S cells, with a specific activity of ≥ 5 x 10^8 units/mg (EC50 ≤ 2 pg/mL); in a representative assay the EC50 was 0.81 pg/mL, and 2 μg were resolved by SDS-PAGE. The endotoxin level is ≤ 0.2 EU/μg protein as determined by kinetic Limulus amebocyte lysate (LAL) analysis. The lyophilized product should be stored at -80 °C and is stable as supplied for 12 months from the date of receipt. Before opening, the vial should be centrifuged and reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; vortexing and repeated freeze-thaw cycles should be avoided.
