Description

Interferon-gamma (IFN-γ), also known as type II interferon, is a cytokine produced by T and NK cells and, in smaller amounts, by dendritic cells and macrophages. It is regulated by cytokines such as IL-12 and IL-18 that are secreted in response to infection. IFN-γ binds to a receptor complex and initiates signal transduction via the JAK/STAT pathway, leading to transcription and activation of genes controlling diverse immunological functions. It stimulates the antimicrobial and anti-tumor activity of macrophages, NK cells, and neutrophils by promoting microbial effector functions such as production of reactive oxygen species, NO intermediates, and complement. IFN-γ enhances MHC class I and II expression in dendritic cells and mononuclear phagocytes, stimulates B cell survival and growth, redirects B cells from proliferation toward differentiation, favors Th1 over Th2 development, and stimulates monocyte differentiation and function.

STEMCELL Technologies supplies Mouse Recombinant IFN-gamma (alternative names: Interferon gamma, Type II interferon), expressed in E. coli and lyophilized from a sterile-filtered aqueous solution containing sodium phosphate and sodium chloride at pH 7.5. The predicted molecular mass is 15.7 kDa. Specific activity is ≥ 1 x 10^6 units/mg (EC50 ≤ 1 ng/mL) as determined in a viral challenge assay using encephalomyocarditis (EMC) virus on L929 cells. Purity is ≥ 95%. Endotoxin level is ≤ 0.1 EU/μg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis. The product should be stored at -20 to -80°C and is stable as supplied for 12 months from date of receipt. For use, the vial and sterile water are brought to room temperature (15 - 25°C) and the product is reconstituted to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing. In a representative biological activity assay with L929 cells exposed to EMC virus in a cytopathic effect (CPE) assay, the EC50 was 0.22 ng/mL and the specific activity normalized to an internal standard was 4.3 x 10^7 units/mg.
