Description

A recombinant mouse cytokine from STEMCELL Technologies, expressed in CHO cells and supplied as a lyophilized powder following dialysis against phosphate-buffered saline. Granulocyte-macrophage colony-stimulating factor (GM-CSF) promotes the proliferation and differentiation of hematopoietic progenitor cells and supports the generation of neutrophils, eosinophils, and macrophages. In synergy with cytokines such as stem cell factor, IL-3, erythropoietin, and thrombopoietin, it also stimulates erythroid and megakaryocyte progenitor cells. GM-CSF is produced by multiple cell types, including stromal cells, Paneth cells, macrophages, dendritic cells, endothelial cells, smooth muscle cells, fibroblasts, chondrocytes, and Th1 and Th17 cells, and drives the development of dendritic cells that ingest, process, and present antigens to the immune system. The receptor for GM-CSF comprises a cytokine-specific α subunit (GMRα; CD116) and a common βc subunit (CD131) shared with the IL-3 and IL-5 receptors.

The product has a predicted molecular mass of 14.2 kDa (UniProt accession Q14AD9) and is reported to be species-specific. Specific activity is ≥ 2.0 x 10^7 units/mg as determined by a cell proliferation assay using FDC-P1 cells, with an EC50 of ≤ 50 pg/mL. Purity is ≥ 95%, and the endotoxin level is ≤ 0.2 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. The product should be stored at -80°C and is stable as supplied for 12 months from the date of receipt. Before opening, the vial should be centrifuged and the contents reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing; as a general guide, reconstituted material should not be stored at 2 to 8°C for more than 1 week or at -20°C for more than 2 months, and repeated freeze-thaw cycles should be avoided.

According to the supplier's safety data sheet, the product is intended as a laboratory reagent and is not classified as a hazardous substance or mixture under the Globally Harmonized System (GHS) or the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200).
