STEMCELL Technologies offers Mouse Recombinant FGF-21, a growth factor that promotes insulin-independent glucose uptake, lipid accumulation and insulin production, and inhibits glucose-mediated glucagon release. The biological activity of the protein was tested by its ability to promote the proliferation of NIH-3T3 cells in the presence of 1.25 μg/mL mouse Klotho and 10 μg/mL heparin, with cell proliferation measured using a fluorometric assay method. The EC50, defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum, is less than 0.5 μg/mL. Purity was assessed by resolving 2 μg of the protein with SDS-PAGE under reducing and non-reducing conditions, visualized by Coomassie Blue staining. The product is designed for use as part of a workflow involving serum-free medium for the differentiation of human ES and iPS cells to pancreatic progenitor cells, alongside related growth factors such as basic fibroblast growth factor, epidermal growth factor, and fibroblast growth factor 8B.
