Description

From STEMCELL Technologies, an animal component-free recombinant cytokine preparation of mouse Receptor Activator of Nuclear Factor Kappa-B Ligand (RANKL; also known as CD254, TNFSF11, OPGL, ODF, TRANCE, and OPTB2). RANKL is a type II homotrimeric transmembrane member of the tumor necrosis factor (TNF) superfamily that exists in both membrane-bound and secreted forms. It binds the RANK receptor and activates the AKT signaling pathway, and is also bound by osteoprotegerin (OPG) in a competitive interaction. The protein, expressed in E. coli, is relevant to studies of osteoclastogenesis and T-cell activation.

Prepared from a sterile-filtered aqueous solution containing 10 mM sodium phosphate and 50 mM sodium chloride at pH 7.5, the protein (accession O35235, residues Pro143–Asp316) has a predicted molecular mass of 19.5 kDa as a monomer. Specifications include ≥ 95% purity, an endotoxin level ≤ 1.0 EU/μg protein by kinetic Limulus amebocyte lysate (LAL) analysis, and activity with an EC50 ≤ 50 ng/mL as determined using RAW-Blue cells.

Store at -20 °C to -80 °C; the lyophilized product is stable as supplied for 12 months from the date of receipt. Centrifuge the vial before opening and reconstitute in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing. The effect of stock-solution storage on performance should be tested for each application, and repeated freeze-thaw cycles should be avoided. For research use only; not intended for human or animal diagnostic or therapeutic uses.
