Ms RANKL, ACF, 100 _g

A recombinant form of the receptor activator of nuclear factor kappa-B ligand (RANKL) is supplied by STEMCELL Technologies as an animal component-free, lyophilized preparation intended for research use. RANKL is a member of the tumor necrosis factor (TNF) superfamily and exists as a type II homotrimeric transmembrane protein in both membrane-bound and secreted forms.

Binding of the cytokine to its receptor, RANK, activates the AKT signaling pathway, and the related molecule osteoprotegerin (OPG) can also bind RANKL in competition with RANKL–RANK binding. RANKL is implicated in osteoclastogenesis and T cell activation.

The mouse sequence (UniProt accession O35235, residues Pro143–Asp316) is produced recombinantly in E. coli, with a predicted monomer molecular mass of 19.5 kDa. The product is lyophilized from a sterile-filtered aqueous solution containing 10 mM sodium phosphate and 50 mM sodium chloride at pH 7.5. Activity is measured using RAW-Blue™ cells, with a specification EC50 of ≤ 50 ng/mL; an example lot value was 11.3 ng/mL. Purity is ≥ 95% and endotoxin level is ≤ 1.0 EU/μg protein by kinetic LAL analysis. The product is stored at -20°C to -80°C and is stated to be stable for 12 months from the date of receipt when supplied.
