The QIAGEN miRCURY Probe PCR Kit delivers a high-performance master mix formulated for probe-based quantification of miRNA, used together with miRCURY LNA miRNA Probe PCR Assays and Panels. The chemistry is built around LNA technology: LNA-enhanced primers and probe have higher Tm, which boosts reaction stringency and enables discrimination between miRNAs that differ by a single nucleotide regardless of AU content. According to QIAGEN, the system allows quantification down to 1 copy of a target miRNA without pre-amplification.

The kit provides the QuantiNova Probe Master Mix and the miRCURY Probe PCR Universal Reverse Primer. Detection relies on a quencher-labeled hydrolysis probe, with miRNA-specific forward primers designed against mature miRNAs using sequence information from miRBase. The mix includes a hot-start DNA polymerase and an optimized buffer system intended to maximize sensitivity and minimize background, and the QuantiNova hot-start mechanism is designed to deliver reproducible results between technical and biological replicates. The mix is optimized for any real-time cycler and supports room-temperature setup and delayed runs for greater workflow flexibility and automation.

The protocol is short, taking RNA to data in 2 hours, with a reduced number of pipetting steps to minimize technical variation. Reverse transcription is performed separately with the miRCURY LNA RT Kit, and the resulting cDNA can be carried across the broader miRCURY LNA miRNA Probe PCR System. ISO-compliant assay manufacturing is intended to contribute to high day-to-day and site-to-site reproducibility.
