Description

QIAGEN offers a high-potency LNA inhibitor for robust miRNA silencing in challenging contexts, including difficult-to-transfect cell lines and tissues, enabling assessment of cellular phenotypes in miRNA loss-of-function studies. Tm-normalized LNA chemistry creates inhibitors with the same high efficacy regardless of the GC content of their miRNA targets, which can vary from 5–95%. The combination of LNA and phosphorothioate modifications dramatically improves stability against enzymatic degradation, giving the Power Inhibitors significantly better efficacy than regular inhibitors, with sub-nanomolar potency under optimal transfection conditions. Because of their high potency, the inhibitors can be added directly to cell cultures without the need for transfection reagents, in a process known as gymnosis.

The inhibitors are antisense oligonucleotides with perfect sequence complementarity to their targets, sequestering the target miRNA in highly stable heteroduplexes to prevent interaction with normal cellular partners. LNA positioning is arranged so that inhibitor/RNA duplexes are not recognized as substrates by RNase H, minimizing off-target effects on mRNA and lncRNA stability. Predesigned inhibitors are available for all known human, mouse and rat miRNAs, covering a large proportion of vertebrate and invertebrate miRNAs, and are also offered with fluorescein (6-FAM) labels. The product is supplied desalted and dried down in tubes.

For handling, the inhibitor should be stored at –20 °C, reconstituted in nuclease-free water, and prepared as aliquots to avoid freeze–thaw cycles.
