miRCURY LNA miRNA Inhibitor (5)

For loss-of-function studies of mature microRNAs, QIAGEN offers the miRCURY LNA miRNA Inhibitor, an LNA-enhanced antisense oligonucleotide that knocks down specific miRNAs in cell-based and in vivo experiments. Locked Nucleic Acid chemistry raises duplex thermal stability and nuclease resistance, increasing binding affinity for the target miRNA and producing stronger knockdown than unmodified DNA antagomirs at low nanomolar concentrations.

Each oligonucleotide is synthesized against a user-selected miRNA sequence, for example hsa-miR-21 or mmu-miR-122, enabling complementarity with miRNA mimic overexpression work. Delivery uses standard transfection reagents, and successful knockdown of the targeted miRNA is verified by RT-qPCR or small RNA sequencing. Typical working concentrations fall in the 25 to 100 nM range for cell culture, with dose-response optimization recommended to identify the lowest effective dose.

Research applications include target validation for predicted miRNA-mRNA interactions, investigation of miRNA roles in differentiation, oncology, metabolism, and inflammation, and confirmation of hits identified by sequencing or microarray screens. The inhibitor is supplied as a single-use vial format, shipped lyophilized or in solution, and is intended for research use only.
