These antisense oligonucleotides have perfect sequence complementarity to their targets and, when introduced into cells, sequester the target miRNA in highly stable heteroduplexes that prevent the miRNA from hybridizing with its normal cellular interaction partners. The Tm-normalized LNA design ensures consistent high efficacy regardless of the GC content of the target miRNA, enabling miRNA functional studies on sequences not represented in miRBase.

The combination of LNA and phosphorothioate modifications dramatically improves stability against enzymatic degradation, so efficacy is significantly better than with regular inhibitors, and enables unassisted "naked" delivery (gymnosis) in serum-containing medium without transfection reagents. LNA positioning ensures that inhibitor/RNA duplexes are not recognized as substrates by RNase H, minimizing off-target effects on mRNA and lncRNA stability, and limiting effects on translation of mRNAs containing complementary sequences in the open reading frame.
