Description

From QIAGEN, this kit is intended for molecular biology applications involving the enrichment of exosomes and other extracellular vesicles from cell culture supernatants, urine, or cerebrospinal fluid (CSF). It is shipped at ambient temperature; all solutions should be kept tightly sealed and stored protected from light at 2–8°C, where they remain stable for at least 6 months in unopened containers.

The isolation procedure relies on a precipitation-based principle that diminishes the hydration of subcellular particles, allowing even particles smaller than 100 nm to be precipitated with a low-speed centrifugation step. Exosomes are cell-derived membranous particles with a size of 20–120 nm. The kit precipitates exosomes, microvesicles and, to some degree, larger protein complexes.

The protocol supports a recommended starting volume of 1–10 mL (maximum 10 mL) and is scalable within that range by adjusting the Precipitation Buffer volume. For best results, the manufacturer recommends urine samples of 2–10 mL, CSF samples of 1 mL, and cell-conditioned media of 1–10 mL. Add 0.4 volumes of Precipitation Buffer for every 1 volume of sample, then incubate at 2–8°C for 60 min. The mixture is centrifuged at 20°C before the pellet is resuspended in ≥100 µL Resuspension Buffer. Ten isolations can be completed in less than 120 minutes. For downstream RNA isolation, the kit is compatible with the miRNeasy Micro Kit or miRNeasy Serum/Plasma Kit. These products are not intended for the diagnosis, prevention or treatment of a disease.
