MinElute PCR Purification Kit (50)

This silica-membrane-based kit from QIAGEN is designed for the purification and cleanup of PCR products ranging from 70 bp to 4 kb in size. It delivers highly concentrated DNA with a binding capacity of 5 µg and allows elution in volumes as small as 10 µl. The kit provides spin columns, buffers, and collection tubes, and is intended for the recovery of oligonucleotides and dsDNA while removing short oligonucleotides (under 10mers and under 40mers) as well as dye terminator proteins.

Workflow follows a simple bind-wash-elute procedure: binding buffer is added to the PCR sample and the mixture applied to the MinElute spin column, where nucleic acids adsorb to the silica membrane under high-salt conditions. An optional pH indicator in the binding buffer helps determine the optimal pH for DNA binding. After washing, DNA is eluted in a small volume of low-salt buffer or water, ready for downstream applications. GelPilot Loading Dye is included, containing three tracking dyes — xylene cyanol, bromophenol blue, and orange G — to assist with agarose gel analysis.

The spin columns can be processed using a conventional table-top microcentrifuge or any vacuum manifold with luer connectors such as the QIAvac 24 Plus, and the procedure is also fully automatable on the QIAcube Connect. Spin columns should be stored at 2–8 °C upon arrival; short-term storage for up to 4 weeks at room temperature (15–25 °C) does not affect performance. For DNA fragments larger than 4 kb, purification with the QIAquick PCR Purification Kit is recommended.
