The dye penetrates and tints the surrounding collagen matrix, creating a light blue field that contrasts sharply with dark, compact CFU-Mk clusters, so technicians can count true colonies under an inverted microscope without confusion from cellular debris. For best results, aliquot the stain into smaller working volumes to minimize freeze-thaw cycles, refrigerate between uses, and protect the bottle from light to preserve dye performance across multiple staining sessions.
