A ready-to-use density gradient medium packaged as six 500 mL bottles, this STEMCELL Technologies reagent enables rapid isolation of mononuclear cells from whole blood through a single centrifugation step. Formulated with sodium diatrizoate and a polysaccharide polymer to deliver a working density of 1.077 g/mL, the solution is applied in immunology, transfusion medicine, and cell therapy workflows where clean lymphocyte and monocyte preparations are required. Standard protocols layer diluted peripheral blood, bone marrow, or cord blood over the medium and centrifuge at 800 x g for 20 minutes at room temperature; viable mononuclear cells collect as a distinct band at the interface while erythrocytes and granulocytes sediment to the pellet. Researchers use the resulting fraction for flow cytometry analysis, in vitro stimulation assays, and downstream cell culture expansion. Each bottle provides enough volume for dozens of standard separations on a benchtop centrifuge with swinging-bucket rotors, and the reagent remains stable at room temperature when protected from light, simplifying storage on the lab bench between sessions.
