Lymphoprep, 500mL

A density gradient separation medium supplied in a 500 mL bottle, this ready-to-use formulation combines sodium metrizoate with a polysaccharide and is adjusted to a working density of 1.077 g/mL for isolating viable mononuclear cells from peripheral blood, bone marrow, or umbilical cord blood.

Anticoagulated blood diluted 1:1 with balanced salt solution is layered gently over the medium and centrifuged at 800 x g for 20 minutes at 20 °C with the brake off. Lymphocytes, monocytes, and other mononuclear cells collect as a distinct white interface above the erythrocyte and granulocyte pellet, while platelets remain in the upper plasma layer. The harvested band is washed twice in buffer before being carried into culture, cryopreservation, or analytical work.

One 500 mL bottle supports roughly 25 to 30 standard 15 mL tube separations. The medium is sterile-filtered and endotoxin-tested, and is routinely used as the first step in flow cytometry workflows, in vitro stimulation assays, cell culture seeding, and the banking of working PBMC stocks for longitudinal studies. Store upright at 18 to 20 °C, protected from light; do not freeze. Shelf life is 24 months from manufacture when the bottle is unopened. Supplied by STEMCELL Technologies for research use only.
