Kartogenin, 5 mg

Supplied by STEMCELL Technologies as a crystalline solid, kartogenin is a small-molecule RUNX1 transcriptional activator. Its chemical name is 2-[(4-phenylphenyl)carbamoyl]benzoic acid (CAS 4727-31-5), with molecular formula C₂₀H₁₅NO₃, molecular weight 317.3 g/mol, and reported purity ≥ 98%. The compound induces chondrogenesis by binding the actin-binding protein filamin A, which disrupts filamin A's interaction with the transcription factor core-binding factor β subunit (CBFβ). Once dissociated from filamin A, CBFβ translocates to the nucleus and forms a transcriptional complex with the runt-related transcription factor RUNX1, enabling chondrocyte differentiation.

Published applications include promoting differentiation of human bone marrow mesenchymal stem cells into chondrocytes, promoting cartilage formation and repair in mouse models of osteoarthritis or when injected into mouse tendon–bone junctions, and promoting type-III collagen synthesis in human dermal fibroblasts in vitro and in the dermis of mice through activation of the SMAD4/SMAD5 pathway. For use as a cell culture supplement, stock solution should be diluted into culture medium immediately before use, with final DMSO concentration kept below 0.1% to avoid potential cell toxicity.

Solubility in DMSO is reported at ≤ 90 mM, with ~0.5 mg/mL in a 1:1 solution of DMSO:PBS (pH 7.2) and ~30 mg/mL in DMSO and DMF. The product is stable at -20°C as supplied, should be protected from prolonged exposure to light, and is stable as supplied for 12 months from date of receipt. According to the manufacturer's Safety Data Sheet, the compound is classified as acutely toxic by ingestion (oral ATEmix 500.00 mg/kg) and as a serious eye irritant, and is described as very toxic to aquatic life with long lasting effects.
